چکیده

The diversity of diazotrophs was studied in the sediment of mangrove forests (Twin Cays, Belize) subjected to a long-term fertilization with nitrogen and phosphorus. Terminal Restriction Fragment Length Polymorphism (TRFLP) and cloning of PCR-amplified nifH genes were combined via in silico analysis to assign clones to TRFLP-nifH phylotypes, as well as to characterize the occurrence of phylotypes in response to environmental conditions. Results indicated that mangrove sediments from Belize harbor a unique diazotrophic community with a low metabolic diversity dominated by sulfate reducers. The variability of potential nitrogen-fixing sulfate reducers was explained by several environmental parameters, primarily by the abundance of dead roots in the sediments, and the concentration of H2S in the pore-waters. This study describes the complexity of microbial communities within the mangrove sediments with specific functional groups varying along environmental gradients. OPEN ACCESS J. Mar. Sci. Anthropogenic impact on diazotrophic diversity in the mangrove rhizosphere revealed by nifH pyrosequencing

Diazotrophs in the mangrove rhizosphere play a major role in providing new nitrogen to the mangrove ecosystem and their composition and activity are strongly influenced by anthropogenic activity and ecological conditions. In this study, the diversity of the diazotroph communities in the rhizosphere sediment of five tropical mangrove sites with different levels of pollution along the north and s...

Abundance and genetic diversity of nifH gene sequences in anthropogenically affected Brazilian mangrove sediments.

Although mangroves represent ecosystems of global importance, the genetic diversity and abundance of functional genes that are key to their functioning scarcely have been explored. Here, we present a survey based on the nifH gene across transects of sediments of two mangrove systems located along the coast line of São Paulo state (Brazil) which differed by degree of disturbance, i.e., an oil-sp...

Profiling gene expression to distinguish the likely active diazotrophs from a sea of genetic potential in marine sediments

Nitrogen (N) cycling microbial communities in marine sediments are extremely diverse, and it is unknown whether this diversity reflects extensive functional redundancy. Sedimentary denitrifiers remove significant amounts of N from the coastal ocean and diazotrophs are typically regarded as inconsequential. Recently, N fixation has been shown to be a potentially important source of N in estuarin...

Long-term nitrogen and phosphorus fertilization effects on N2 fixation rates and nifH gene community patterns in mangrove sediments

The bioavailability of nutrients in sediments is one of the most important factors controlling ecological and physiological processes in the mangrove ecosystem (Alongi et al. 2002; Feller et al. 2002; Cheeseman & Lovelock 2004; Krauss et al. 2008). Although mangroves are highly productive, they are generally nutrient-deficient, relying on microbial and plant cycling processes for nutrients (Hol...

Assessing Heavy Metals Pollution of the Mangrove Sediments along Tidal Flat of the Mehran River Delta, Persian Gulf

The relationship between the concentration of heavy metals and physicochemical factors was studied in the fine-grained sediments of the tidal section of the Mehran delta where mangrove trees have grown. Surface sediments of the tidal zone of the Mehran delta were sampled. The grain size distribution, calcium carbonate, organic matter contents, pH/Eh, and heavy metals concentration were determin...
